چکیده
The pathogenesis of esophageal squamous cell carcinoma (ESCC) involves both genetic and environmental factors. Previously, we have carried out gene and protein expression profiling of ESCC using DNA microarrays and mass spectrometry-based quantitative proteomics, respectively. These studies resulted in identification of several potential biomarkers of ESCC, some with known reports of differential expression in the scientific literature and others that were novel observations from our studies. We report systematic validation of selected markers from our studies on a larger cohort of cancer tissue sections by immunohistochemical labeling of tissue microarrays. We have validated expression of insulin-like growth factor-binding protein 7 (IGFBP7), stanniocalcin 2 (STC2), inhibin beta A (INHBA) and four and a half LIM domains 1 (FHL1). Immunohistochemical labeling with anti-stanniocalcin 2 antibody demonstrated its overexpression in 132/140 (94%) cases, IGFBP7 showed overexpression in 127/140 (91%) cases and overexpression of INHBA was observed in 62/105 (59%) of ESCC cases. In contrast, FHL1 expression was observed only in 12/143 (8%) of ESCC cases suggesting its possible involvement in tumor suppression. These data suggest that IGFBP7, INHBA, STC2 and FHL1 might play an important role in ESCC tumorigenesis, which can be explored in future studies. Overall, our findings open up new avenues for development of novel therapeutics and/or diagnostic approaches in ESCC.

Production and functional characterization of human insulin-like growth factor 1
Insulin-like growth factor 1 (IGF-1) is a polypeptide hormone produced mainly by the liver in response to the endocrine growth hormone (GH) stimulus. This protein is involved in a wide range of cellular functions, including cellular differentiation, transformation, apoptosis suppression, migration and cell-cycle progression and other metabolic processes. In the current study, human heart cDNA w...
متن کاملThe Effect Of P53 Protein Over-Expression And Its Clinical Features On The Response To Preoperative Chemoradiotherapy Of Esophageal Squamous Cell Carcinoma
Background and Objective: P53 is a suppressive gene that plays a key role in DNA repair and apoptosis. The purpose of this study was to investigate the effect of P53 protein over-expression and some clinicopathological factors on the esophageal squamous cell carcinoma (SCC) response to neoadjuvant chemoradiotherapy. Patients and Methods: In this retrospective cohort study, 44 patients with loc...
متن کاملInduction of Apoptosis by a Combination of 2-Deoxyglucose and Metformin in Esophageal Squamous Cell Carcinoma by Targeting Cancer Cell Metabolism
Background: Both mitochondrial dysfunction and aerobic glycolysis are signs of growing aggressive cancer. If altered metabolism of cancer cell is intended, using the glycolysis inhibitor (2-deoxyglucose (2DG)) would be a viable therapeutic method. The AMP-activated protein kinase (AMPK), as a metabolic sensor, could be activated with metformin and it can also launch a p53-dependent metabolic ch...
متن کاملEvaluation of Cyclin D1 Expression in Esophageal Squamous Cell Carcinoma and its Effect on Response Rate to Neo- adjuvant Chemoradiotherapy
Background and Objective: Esophageal cancer especially squamous cell carcinoma (SCC) is one of the most common gastro intestinal malignancies in north part of Iran (Khorasan). The standard treatment for esophageal cancer is surgical resection, but its outcome remains poor. Then, the oncologists try to treat this cancer with sandwich protocols especially neo-adjuvant chemo-radiotherapy. Sever...
متن کاملP53 and Ki67 protein expression in ocular squamous cell carcinomas of dairy cattle
Role of the protein overexpression is very important in the pathogenesis, diagnosis and prognosis of neoplasia. Cases of overexpression of p53 and Ki67 proteins were investigated in 19 ocular squamous cell carcinomas (OSCC) in dairy cows. Microscopically, insix cases the tumors were carcinoma in situ (noninvasive carcinoma, G0), and in the remaining 13 cases the tumors were invasive carcinoma. ...
